چکیده

Polyneuritis equi is an immune-mediated neurodegenerative condition in horses that is related to circulating demyelinating antibodies against equine myelin basic protein 2 (MP2). The present study examined the presence of circulating demyelinating antibodies against neuritogenic peptides of MP2 in sera from horses suspected of equine protozoal encephalomyelitis (EPM), a neurodegenerative condition in horses that may be immune-mediated. The goals of this study were to develop serum ELISA tests that may identify neuroinflammatory conditions in horses with EPM and indirectly relate the pathogenesis of inflammation to IL6 by serum C-reactive protein (CRP) concentration. Circulating antibodies against MPP, recombinant equine myelin P2 protein (rMP2), and CRP were assayed in 52 neurologically abnormal horses by ELISA in this retrospective study. We detected antibodies against MPP (40%), rMP2 (33%), and CRP (65%) in these horses that had a presumptive diagnosis of EPM. No cross-reactivity between the antigens was observed. An evaluation of the agreement between the assays (McNemar’s test) suggests as CRP values increase, the likelihood of a positive MPP ELISA also increases. Clinical signs of EPM may be due to an immune-mediated polyneuropathy that involves complex in vivo interactions with the IL6 pathway because MPP antibodies and elevated CRP concentrations were detected in some horses with S. neurona

Serum Antibodies Against a Reactive Site of Equine Myelin Protein 2 Linked to Polyneuritis Equi Found in Horses Diagnosed with EPM

Equine protozoal myeloencephalitis (EPM) is the most commonly diagnosed infectious neuro-degenerative condition in horses and neuroinflammation is present in all definitively diagnosed cases. Historically EPM is difficult to treat. Polyneuritis equi (cauda equina neuritis) is a rarely recognized noninfectious, chronic relapsing, peripheral neurodegenerative disorder in horses. We propose that p...

Cellular hypersensitivity to basic myelin (P2) protein in the Guillain-Barré syndrome.

Lymphocytes of 29 subjects were assayed for MIF production in response to P2 peripheral nerve protein, crude human peripheral nerve and human central nervous system Al basic myelin protein. Seven were performed in normal control subjects, 12 in Guillain-Barre patients (GB), 5 with other polyneuropathies and 5 in patients with multiple sclerosis (MS). Only GB patients with acute illness produced...

Immune responses to retinal autoantigens and peptides in equine recurrent uveitis.

PURPOSE To test the hypothesis that autoimmune mechanisms are involved in horses in which equine recurrent uveitis (ERU) develops spontaneously. METHODS Material obtained from horses treated for spontaneous disease by therapeutic routine vitrectomy was analyzed for total IgG content and IgG specific for S-Antigen (S-Ag) and interphotoreceptor retinoid-binding protein (IRBP). The cellular infi...

Equine herpes virus type 1 (EHV-1) and 4 (EHV-4) infections in horses and donkeys in northeastern Turkey

The herpesviruses infections in equides are caused by five different serotypes of viruses, belonging to family Herpesviridae. The goal of this study was to conduct a seroepidemiological investigation of equine herpesvirus type 1 (EHV-1) and type 4 (EHV-4) in horses and donkeys raised in two provinces and their villages in northeastern Turkey. A total of 666 samples from 423 horses and 243 donke...

Detection and quantification of antibodies to the extracellular domain of P0 during experimental allergic neuritis.

Quantification of the peripheral nerve myelin glycoprotein P0 and antibodies to P0 is difficult due to insolubility of P0 in physiological solutions. We have overcome this problem by using the water-soluble recombinant form of the extracellular domain of P0 (P0-ED) and describe newly developed assays which allow detection and quantitation of P0 and antibodies to P0, in serum and cerebrospinal f...
